Cele mai rezistente bacterii \u00een spitalele din Rom\u00e2nia sunt reprezentate de tulpini Gram negative non-fermentative de <em>P. aeruginosa<\/em>. Scopul acestui proiect este de a investiga efectul nanoparticulelor de magnetit\u0103 bioactive functionalizate cu compusi naturali obtinuti din plante asupra virulen\u021bei \u0219i persisten\u021bei unor tulpini de laborator \u0219i izolate clinice, rezistente de <em>P. aeruginosa<\/em>. Abordarea noastr\u0103 inovatoare utilizeaz\u0103 concentra\u021bii subinhibitorii de compu\u0219i deriva\u021bi din plante, livrate controlat, pentru a modula comportamentul \u0219i virulen\u021ba microorganismelor, nu uciderea acestora, \u00een scopul de a limita dezvoltarea \u0219i r\u0103sp\u00e2ndirea de mutante rezistente. Cunoa\u0219terea mecanismelor intime de ac\u021biune ale acestor nanosisteme bioactive cu efect antimicrobian dovedit asupra celulelor de <em>P. aeruginosa<\/em> va \u00eembun\u0103t\u0103\u021bi \u00een mod semnificativ cunoasterea si va sustine dezvoltarea unor metode alternative de preventie si tratament a infectiilor cu acest agent patogen. Proiectul va avea un impact deosebit nu numai pe domeniul stiintific, dar, de asemenea, \u0219i pe sectorul socio-economic, deoarece \u00ee\u0219i propune s\u0103 dezvolte o metod\u0103 alternativ\u0103 de a rezolva sau de a atenua o problem\u0103 global\u0103, a\u0219a cum este rezisten\u021ba la antibiotice.<\/p>\n<p><strong>Abstract<\/strong> (English)<\/p>\n<p style=\"text-align: justify;\">Nowadays, about 70 percent of the bacteria that cause infections in hospitals but also in communities are resistant to at least one of the drugs most commonly used for treatment, causing infections that often fail to respond to standard therapy. Most resistant microorganisms in Romanian hospitals are the Gram negative non-fermenting strains of <em>P. aeruginosa<\/em>. The aim of this project is to investigate the effect of bioactive magnetite nanoparticles functionalized with plant-derived compounds on the virulence and persistence of some laboratory and clinically isolated, resistant <em>P. aeruginosa<\/em> strains. Our innovative approach is using subinhibitory concentrations of plant-derived compounds to modulate microbial behavior and virulence, rather than induce their killing, in order to limit the development and spread of resistant mutants, since strategies that interfere with bacterial population fitness are selecting resistant mutants. Knowing the intimate mechanisms of action of these bioactive nanoformulations with proved antimicrobial effect on the <em>P. aeruginosa<\/em> cells will significantly improve knowledge and offer further perspectives in developing alternative preventing and treatment approaches of this pathogen. This project is expected to have a great potential not only on the research field but also on the socio-economic sector, since it aims to develop an alternative method to solve or attenuate a global problem, as is the antimicrobial resistance.<\/p>\n<p><strong>Obiectivul<\/strong> <strong>proiectului<\/strong><\/p>\n<p style=\"text-align: justify;\">Scopul acestui proiect este de a investiga efectul nanoparticulelor de magnetit\u0103 bioactive functionalizate cu compusi naturali obtinuti din plante asupra virulen\u021bei \u0219i persisten\u021bei unor tulpini de laborator \u0219i izolate clinice, rezistente de <em>P. aeruginosa<\/em>.<\/p>\n<p><strong>Rezultatele estimate\u00a0\u00a0 <\/strong><\/p>\n<p style=\"text-align: justify;\">Proiectul propus va aduce un aport semnificativ asupra cunoasterii interac\u021biunilor specifice dintre nanoparticulele de magnetita functionalizate cu compusi antimicrobieni derivati din plante asupra oportunistului <em>P. aeruginosa<\/em>, at\u00e2t la nivel fenotipic cat si molecular. Acest proiect \u00ee\u0219i propune s\u0103 sprijine \u0219i s\u0103 extind\u0103 direc\u021biile recente de cercetare dezvoltate de grupul nostru de cercetare pe domeniul nanoparticulelor bioactive cu aplicatii in terapia antimicrobian\u0103. In plus, acest proiect deschide noi direc\u021bii de cercetare pentru dezvoltarea unor terapii antimicrobiene alternative personalizate, \u00een func\u021bie de particularit\u0103\u021bile agentului patogen incriminat. Proiectul sus\u021bine o direc\u021bie de cercetare recent dezvoltata ce vizeaza manipularea infec\u021biilor severe prin modularea virulen\u021bei patogenilor dificil de eradicat. Sectorul tehnologic va beneficia semnificativ de realizarea acestui proiect, deoarece nanoparticule specifice vor fi proiectate pentru a furniza, stabiliza, poten\u021ba \u0219i asigura o eliberare adecvat\u0103 a substantelor antimicrobiene naturale. Proiectul va utiliza toate mijloacele accesibile (conferin\u021be, publicatii pe domeniu etc) pentru cre\u0219terea gradului de con\u0219tientizare cu privire la acest domeniu. Proiectul va avea un impact deosebit nu numai pe domeniul stiintific, dar, de asemenea, \u0219i pe sectorul socio-economic, deoarece \u00ee\u0219i propune s\u0103 dezvolte o metod\u0103 alternativ\u0103 de a rezolva sau de a atenua o problem\u0103 global\u0103, a\u0219a cum este rezisten\u021ba la antibiotice. Studiile arata ca pentru pacien\u021bii cu infec\u021bii rezistente \u0219i persistente cheltuielile de spitalizare cresc cu $US296 milioane pana la $US2.3 miliarde de dolari anual. Cu toate ca devin tot mai inficiente, antibioticele reprezint\u0103 a treia cea mai vanduta clas\u0103 de medicamente, cu o pia\u021b\u0103 anual\u0103 \u00eentre 7 \u0219i 22 miliarde de dolari numai \u00een SUA. Terapiile alternative antimicrobiene pe baz\u0103 de produse naturale ar putea s\u0103 sc\u0103d\u0103 semnificativ nu numai riscul de a dezvolta antibio-rezisten\u021b\u0103, dar, de asemenea, costurile associate terapiei, precum \u0219i efectele nocive asociate, manifestate asupra mediului si pacientului. Antibioticele nu sunt degradate complet \u00een corpul animal \u0219i constituie o sursa semnificativa de poluanti \u00een sol \u0219i ape. In plus, aceste medicamente au efecte secundare grave asupra pacien\u021bilor, \u00een special atunci c\u00e2nd sunt utilizate in cantit\u0103\u021bi mari pentru tratarea infec\u021biilor cauzate de bacterii patogene rezisten\u021be, precum <em>P. aeruginosa<\/em>. Produsele naturale sunt folosite de secole \u00een medicina tradi\u021bionala, cu efecte secundare nesemnificative. Luate \u00eempreun\u0103, aceste aspect recomand\u0103 proiectul acesta ca o abordare \u0219tiin\u021bific\u0103 \u0219i tehnologic\u0103 inovatoare \u0219i eficient echilibrat\u0103 \u00een manipularea infec\u021biilor grave, cauzate de un agent patogen dificil de controlat, P. aeruginosa.<\/p>\n<p style=\"text-align: justify;\">Pe parcursul desfasurarii acestui proiect se estimeaza scrierea \u0219i publicarea a cel pu\u021bin 3 lucr\u0103ri lucr\u0103ri \u0219tiin\u021bifice in reviste de prestigiu indexate ISI-Web of knowledge, a cel putin 2 capitole de carte\/ sau articole in reviste cotate BDI si sustinerea a cel putin 3 comunicari orale sau de tip poster la conferinte internationale.
